The human whole blood samples containing four drugs (amitriptyline, chlorimipramine, imipramine and trimipramine) were heated at 100 °C in a septum-capped vial in the presence of NaOH solution, and a polydimethylsiloxane-coated SPME fiber was exposed to the headspace of the vial to allow adsorption of the drugs before capillary gas chromatography (GC) with flame ionization detection. The headspace SPME-GC gave intense peaks for each drugs; only a few small background noises appeared. Recovery of the four drugs with use of the headspace SPME-GC were 5.8-12.9 %. Reproducibility for headspace SPME-GC data was excellent for whole blood samples. The calibration curves for the drugs showed linearity in the range of 31-1000 ng/0.5 ml for whole blood. The detection limits of each drugs were 16-25 ng/0.5 ml for whole blood.
