These systems express the major histocompatibility complex class I (MHCI), a neuro-immune molecule recently identified to be present in the neurons and glial cells of the brain. The MHCI molecules regulate neural connectivity and plasticity in the developing as well as adult brain. Here we report that mice in which functional MHCI has been lacked exhibit hyperactivity, motor impulsiveness and attention deficits that are three major symptoms of ADHD. They also show alterations in the gene expression of dopamine synthetic enzyme, transporter and receptor subtypes in the cell body and terminal areas of the midbrain dopamine systems. Moreover, their behavioral phenotypes are improved, although not completely reversed, by methylphenidate, an ADHD medicine.
