CA125の精製は0.6M過塩素酸処理, Sepharose CL-4B, 6M urea加熱処理後Sepharose 6Bによるゲルろ過, およびOC125 affinityカラムにより行った. HOC-I細胞および卵巣癌組織抽出液より精製されたCA125抗原の分子量は1,000KDa以上を呈した. 一方, HOC-I細胞培養上清および卵巣癌患者血清からは分子量110KDaから400KDaに分布するCA125が精製された. CA125分子が他のglycoconjugatesと複合体を形成していることを確認するために, 各種抗体 (OC125, NS19-9, TKH-2, B72.3, CC49, MA54, MA61, PC47H) を96穴ELISAプレートに固相し, probeとしてbiotin標識OC125を用いてdouble-determinant enzyme-linked immunosorbent assay (ELISA)を行った.
