A simple method, for the extraction of four tricyclic antidepressants from human urine, using the headspace solid-phase microextraction (SPME) is presented. After heating a vial containing urine sample and the drugs at 100℃ in the presence of NaOH solution, a polydimethylsiloxane-coated SPME fiber was exposed in the headspace of the vial to allow adsorption of the drugs. The fiber needle was then injected into a capillary gas chromatography (GC) port. The headspace SPME-GC gave intense peaks for each drugs; only a few small background noises appeared. Recovery of the four drugs with use of the headspace SPME-GC were 44-81 %. The calibration curves showed linearity in the range of 0.05-2μg/ml. The detection limits of each drugs were 24-38 ng/ml. The present results on the analysis of tricyclic antidepressants by headspace SPME suggest its applicability to a number of other solid drugs and poisons of middle-sized molecular weights in forensic toxicology . |
